چکیده

Multivariate nonlinear mixed-effects models (MNLMM) have received increasing use due to their flexibility for analyzing multi-outcome longitudinal data following possibly nonlinear profiles. This paper presents and compares five different iterative algorithms for maximum likelihood estimation of the MNLMM. These algorithmic schemes include the penalized nonlinear least squares coupled to the multivariate linear mixed-effects (PNLS-MLME) procedure, Laplacian approximation, the pseudo-data expectation conditional maximization (ECM) algorithm, the Monte Carlo EM algorithm and the importance sampling EM algorithm. When fitting the MNLMM, it is rather difficult to exactly evaluate the observed log-likelihood function in a closed-form expression, because it involves complicated multiple integrals. To address this issue, the corresponding approximations of the observed log-likelihood function under the five algorithms are presented. An expected information matrix of parameters is also provided to calculate the standard errors of model parameters. A comparison of computational performances is investigated through simulation and a real data example from an AIDS clinical study.

Multivariate inhomogeneous diffusion models with covari- ates and mixed effects

Modeling of longitudinal data often requires diffusion models that incorporate overall time-dependent, nonlinear dynamics of multiple components and provide sufficient flexibility for subject-specific modeling. This complexity challenges parameter inference and approximations are inevitable. We propose a method for approximate maximum-likelihood parameter estimation in multivariate time-inhomog...

An approximate likelihood approach to nonlinear mixed effects models via spline approximation

In dealing with parametric nonlinear mixed effects models, intensive numerical integration often makes exact maximum likelihood estimation impractical given the current computing capacity. Algorithms based on linearization, such as the first order method and the conditional first order method, have the potential of producing highly inconsistent estimates, although numerically they are more effi...

A comparison of algorithms for maximum likelihood estimation of Spatial GLM models

In spatial generalized linear mixed models, spatial correlation is assumed by adding normal latent variables to the model. In these models because of the non-Gaussian spatial response and the presence of latent variables the likelihood function cannot usually be given in a closed form, thus the maximum likelihood approach is very challenging. The main purpose of this paper is to introduce two n...

Estimation of parameters in incomplete data models defined by dynamical systems

Parametric incomplete data models defined by ordinary differential equations (ODEs) are widely used in biostatistics to describe biological processes accurately. Their parameters are estimated on approximate models, whose regression functions are evaluated by a numerical integration method. Accurate and efficient estimations of these parameters are critical issues. This paper proposes parameter...

Estimation in nonlinear mixed-effects models using heavy-tailed distributions

Nonlinear mixed–effects models are very useful to analyze repeated measures data and are used in a variety of applications. Normal distributions for random effects and residual errors are usually assumed, but such assumptions make inferences vulnerable to the presence of outliers. In this work, we introduce an extension of a normal nonlinear mixed–effects model considering a subclass of ellipti...
